Protein and Glycoprotein Patterns Related to Morphogenesis in Horseradish Tissue Culture
نویسندگان
چکیده
In this study cellular proteins of in vitro grown horseradish (Armoracia lapathifolia Gilib.) tissues of different morphologies, leaves (L), teratoma (TM) and tumor (TN), were separated by one-dimensional (1-DE) and two-dimensional polyacrylamide gel electrophoresis (2-DE) in order to reveal developmentally specific proteins. Moreover, proteins were subjected to analyses by lectin assays to reveal whether the morphological differences were reflected in protein glycosylation. 1-DE glycoprotein pattern showed substantial distinctions between horseradish tissues of different morphologies, which was in contrast to silver stained protein profiles, where only slightly qualitative changes were observed. 2-DE revealed more differences in expression of cellular proteins between examined tissues in comparison to 1-DE, while 2-DE blots confirmed quantitative difference in expression of 43 kDa glycoprotein between investigated tissues. Mass spectrometry analysis identified this protein as a typical horseradish peroxidase. Obtained results imply that the oligosaccharide structures could be important determinants of specificities of horseradish tissue morphology. (doi: 10.5562/cca2078)

A Chick Embryo in-Vitro Model of Knee Morphogenesis
Background: In this feasibility study, a mechanically loaded in-vitro tissue culture model of joint morphogenesis using the isolated lower extremity of the 8 day old chick embryo was developed to assess the effects of mechanical loading on joint morphogenesis. Methods: The developed in-vitro system allows controlled flexion and extension of the chick embryonic knee with a range of motion of ...
متن کاملLectin Histochemical Study of Vasculogenesis During Rat Pituitary Morphogenesis
Objective(s) The aim of this study was to investigate glycoconjugates distribution patterns as well as their changes during the course of pituitary portal vasculogenesis and angiogenesis. Materials and Methods Formalin fixed paraffin sections of 10 to 20 days of Sprague Dawly rat fetuses were processed for histochemical studies using four different horseradish peroxidase (HRP) conjugated lec...
متن کاملSpontaneous Mesenchymal to Epithelial Like Tissue Transition (MET) in a Long Term Human Skin Culture
In an attempt to isolate multipotent stem cells from foreskin in a long-term culture, we encountered an interesting phenomenon which was the conversion of the fibroblast dominant condition to epithelial-like tissue formation. However, the basic mechanism(s) which may be involved in this conversion is not clear. This study was designed to evaluate the cells protein secretion activity and examine...
متن کاملI-28: Role of Mevalonate-Ras Homology (Rho)/Rho-Associated Coiled-Coil-Forming Protein Ki nase-Mediated Signaling Pathway in The Pathogenesis of Endometriosis-Associated Fibrosis
Background: Endometriosis, a disease affecting 3-10% of women of reproductive age, is characterized by the ectopic growth of endometrial glands and stroma surrounded by dense fibrous tissue. Whereas, normal eutopic endometrium shows scarless tissue repair during menstrual cycles, which suggests that the endometriotic tissues have distinct mechanisms of fibrogenesis. During the development of en...
متن کاملDevelopmentally specific soluble and membrane proteins and glycoproteins in Mammillaria gracillis Pfeiff. (Cactaceae) tissue culture
Artificial environmental conditions in the tissue culture influence growth and induce aberration from the characteristic organization pattern. As CAM plants, cacti are particularly susceptible to altered growth environment. In vitro propagated Mammillaria plants spontaneously produce callus which regenerates normal and hyperhydric shoots without the addition of any growth regulator. In order to...
